Effect of Ethanol or Thiamine Administration to Cadmium-Intoxicated Rats on some Essential Metals in Brain Tissue and Serum and some Brain Mitochondrial Enzymes | ||

| Abstract | ||
| This study was conducted on forty male albino rats divided into four equal groups. Group I served as control animals. Group II were injected with cadmium subcutaneously daily for 15 days. Group III were given Cd as in group Il plus ethanol intragastrically daily during the experimental period. Group IV were given Cd as in group II plus thiamine pyrophosphate chloride injected intraperitoneally daily for the experimental period. At the end of the experiment, blood and brain samples were collected. There was significant decrease in serum Zn and Ca in Cd treated group as compared to control group. [p < 0.05] Group II [Cd+ethanol] showed significant decrease in serum Zn and Ca as compared to group II [Cd alone] [p < 0.05]. Group IV [Cd+TPP] exhibited significant increase in serum Zn and Ca as compared to group II [Cd alone] [p < 0.05]. Brain Cu was significantly lower [p < 0.05] while brain Zn was significantly elevated in Cd treated group as compared to control [p < 0.05]. Group IIl [Cd+ethanol] showed significant elevation in brain Zn as compared to Cd treated group [p < .05]. Group IV [Cd+TPP] showed significant increase in brain Cu and a significant reduction in Brain Zn as compared to group II [Cd alone] [p < 0.05] while it exhibited no difference as compared to control group. Brain isocitrate ehydrogenase, a-ketoglutarate dehydrogenase, succinate dehydrogenase and malate dehydrogenase activities were significantly lower in Cd- treated than control group [p < 0.05] and in Cd+ethanol treated group as compared to Cd-treated group [p < 0.05). Group IV [Cd+TPP] illustrated significant elevation in the studied enzyme activities in brain homogenate as compared to Cd-treated group [p < 0.05] as well as the Cd+ethanol treated group. [p < 0.001] Data also show that group IV was not statistically different from control group. It can be concluded that the intervention of Cd with tissue essential metals may play a role in Cd toxicity in rats. Mg is the most stable essential element in case of Cd intoxication. Coadministration of ethanol and Cd intensified the Cd induced changes in rat brain and serum. Thiamine supplementation ameliorates Cd-induced damage both at serum and at brain tissue level. | ||
