| Abstract | ||
| Aim: The current study seeks to evaluate the anticancer role of Cassia alata methanolic leaves extract (CAMLE) in ethyl carbamate-induced lung adenocarcinoma (LUAD) in comparison to the role of Cisplatin (CPL) drug using histological, histochemical, immunohistochemical stains and transmission electron microscopic (TEM) study. Materials and Methods: The rats were divided into four groups of ten rats each: group (1) the control (CONT) group, group (2) the lung adenocarcinoma (LUAD) group in which rats intra-peritoneally injected with 1g/kg ethyl carbamate once weekly for a month, group (3) the LUAD+CAMLE group in which rats orally administered 500 mg/kg CAMLE for the last two months of the experiment and group (4) the LUAD+CPL treatment group, rats intraperitoneally injected 2.5 mg/kg Cisplatin once weekly for the last two months of the experiment. Standard hematoxylin and eosin staining, Masson trichrome staining, Alcian blue-PAS reaction and immunohistochemistry staining for PCNA were performed in addition to terminal bronchiolar Clara cells investigation using transmission electron microscopy. Results: Both light and electron microscopes revealed development of adenocarcinoma inside terminal bronchiole with some histo-pathological changes in LUAD group as abnormal excessive collagen fibers, elevated mucinous content, and increased PCNA positive immunoreactivity while electron microscopy revealed that papillary adenocarcinoma originated from Clara cells in LUAD group. The LUAD+CAMLE treated group showed absence of tumor masses and almost restored normal lung histology with decreased collagen fiber, mucinous content, and PCNA positive immunoreactivity. Moreover, it restored normal ultra-structure of bronchiolar Clara cells. Conclusion: the present study concluded that CAMLE treatment provides a new alternative with less toxicity than Cisplatin drug in lung cancer treatment. therefore, Cassia alata leaves extract should be used as a novel source of anti-cancer compounds fighting lung cancer. | ||
