| Abstract | ||
| Background: Escherichia coli is considered as a major cause of urinary tract infections (UTIs). These bacteria are opportunistic pathogens that have the potential to cause many infections. Objectives: To detect the production of hemolysin by UPEC and investigate the impact of temperature, pH, and glucose on the expression of hemolysin. Methods: 100 midstream urine samples were collected from patients suffering symptoms referred to as UTIs from three hospitals in Baghdad. Biochemical tests and VITEK-2 system were used in the identification of UPEC. The disc diffusion method was employed in the antibiotic sensitivity tests according to CLSI 2022 guidelines. The production of hemolysin was tested on blood agar. Colony PCR was used in the detection of the hlyA gene. RT-PCR was used to determine the hlyA gene expression under different growth conditions of temperature, pH, and glucose concentration. Results: Out of 100 samples, 50 isolates (50%) were E. coli. 94% of the isolates were resistant to Piperacillin, 74% to Aztreonam, 66% to Trimethoprim, 56% to Ciprofloxacin, 54% to Tetracycline, 42% to Amikacin, 38% to Imipenem, 34% to Gentamicin, and 24% to Chloramphenicol. 41 isolates (82%) have the hlyA gene. The hlyA gene expression was increased at 45ºC, pH 7 and glucose concentration at 0.6%. Conclusion: This study showed that UPEC was the predominant bacteria isolated from patients suffering from UTIs.
